Until a few years ago, most strains of Acinetobacter could be killed with a variety of drugs; for those that couldn’t, doctors once relied on broad-spectrum antibiotics known as carbapenems. One drug, Colistin, is the only option that sometimes works, but it comes with a price: it is so incredibly toxic that it can cause kidney damage.

While Gram-negative bacteria do not receive as much attention as methicillin-resistant Staphylococcus aureus (MRSA), some infectious-disease specialists say they could emerge as a bigger threat.  For a combination of business reasons and scientific challenges, the pharmaceuticals industry is pursuing very few drugs for Acinetobacter and other organisms of its type. Meanwhile, the germs are evolving and becoming ever more immune to existing antibiotics.

“These are some of the most antibiotic-resistant bacteria out there, and they can cause all sorts of infections,” says Dr. Gregory Tew of the University of Massachusetts Amherst. Although most infections occur in hospitalized patients, the numbers are quietly escalating in people who are not hospitalized, elderly or immunocompromised. That is why the US Office of Naval Research (ONR) is currently funding new approaches, including the use of defensins.

The ONR recently awarded Dr. Tew and PolyMedix Inc., a development-stage biotechnology company, a $100,000 research contract. The primary goal of the work is to fund investigation of PolyMedix’s novel antimicrobial compounds, the company’s small molecule defensin-mimetics, for activity against bacterial pathogens most relevant to the military, especially Acinetobacter.

In addition to the Navy research contract, in July 2009, PolyMedix, along with the University of Massachusetts Amherst, shared a $6.6 million grant from the National Institutes of Health (NIH) to develop new antibiotics to combat foodborne pathogens and Gram-negative infections.  PolyMedix has also received a $1.6 million research contract from DTRA, the Defense Threat Reduction Agency, and a $100,000 research contract from the US Army, to study its antibiotic compounds for other biodefense applications and bacterial infections. In total, PolyMedix has received nine grants or research contracts to support its research with its defensin-mimetic antibiotic compounds.

PolyMedix’s compounds are non-peptide small molecule drugs that mimic the activity of host-defense proteins. The company’s antibiotic compounds, including PMX-30063, have a mechanism of action distinct from those of current antibiotic drugs, a mechanism that is intended to make bacterial resistance unlikely to develop.
